If I set a passphrase for WPA2, does that mean I have enter the passphrase every time I turn on the router? Otherwise, when does one need to enter it?

Just once, unless you clear the router data by holding in the reset for an extended period.
Pen is right about the router.
As for your PC(s), the first time they connect to the wireless router it/they will ask for the password. Normally there is a little box you can click on that tells the individual PC's to use that password each time to automatically connect to the router at a later time.
If you do not click on the automatic option the PC will ask for the password every time you try to log into the router, usually after a PC reboot.
