Do you know if the Cisco Catalyst (IOS AdvEnterprise) 6500 with Sup720-3B can supply VPLS ?

No, VPLS is not supported on the Catalyst 6500 series. You should look at the 7600 series with the same or better/newer Routing processor. Also remember, you need enhanced services interfaces (ES og SIP-400/600) corefacing.
